Mitosis in onion root tip

College- CMR National PU College, ITPL,Bangalore

Sample- Root tip of onion, grown in lab to study mitosis

Students involved- II PUC

Students of II PUC studied mitosis in onion root tip. In order to observe mitosis, root tips were gently heated with dilute HCl and stained with Acetocarmine. The samples were left undisturbed for sometime and then observed under Foldscope as well as low and high powers of compound microscope.

We could clearly identify prophase, metaphase, anaphase and telophase in many cells using high power of compound microscope (magnified 450 times). But we were not able to identify the phases clearly using foldscope magnification of 140x.

It would be great if we can have higher magnification in foldscope. Is it possible? Please leave a comment if this can be done.

The above picture was captured while observing the sample under high power of compound microscope.
