The methodology of the experiments carried out:
Observation of the transverse section of the stem and root of a dicot plant:
A temporary mount of the transverse section of the stem and root of a dicot plant was prepared as follows:

  1. A thin transverse (cross section) section of the stem or root of the Indian borage (Plectranthus amboinicus ) was removed and was transferred to a watch glass containing the distilled water.
  2. Few drops of the safranin staining solution was then added to the watch glass and stained for 3 minutes.
  3. The specimen was placed on the centre of a microscopic slide and a drop of glycerol was added.
  4. A coverslip was placed over the specimen and any excess water/stain was removed using a blotting paper. The sides of the coverslip were attached to the microscopic slide with an adhesive tape.
  5. The specimen was observed under foldscope.
  6. The foldscope containing the specimen was placed on the stage of the compound microscope and was observed using the microscope’s 10 X objective lens.
