North-East Twinning Program: Hands-on-Demonstration of Foldscope and Its Practical Application in Education-#Indiafoldscopephase 1

The spores on mango kernels were observed under foldscope and we found Aspergillus species. Aspergillus was first catalogued in 1729 by the Italian priest and biologist Pier Antonio Micheli. Aspergillus sp. are widely distributed in nature, it is isolated from a variety of substrates or environments such as humans, air, soil, plants and decaying organic matter. Aspergillus is one of the most important fungal genera in the food spoilage and animal feeds.

Purchased onion from local market of Bangalore, Karnataka was found infested with black mold. It was decided to have a closer look at the black powder like material using foldscope.

 Black mold on onion is commonly caused by a fungus known as
 
 Aspergillus niger.
 
 This fungus is commonly found in soil.

 This saprophytic fungus is generally found on or between the outer scale leaves of the onion bulb. The spore of this fungus spread with the blowing wind and infection occurs on onion surface at high humidity surrounding within 6 to 12 hours.

In our institute garden of Jyothy Institute of Technology (Bangalore), we found bean leaves infected with some disease. On the leaf surface there were dark black spots surrounded by yellow zones.

 It is leaf spot. This fungal disease, caused by
 
 Cercospora
 
 species, occurs primarily on the lower leaves of plants as irregular, tan spots. Severe infection causes excessive leaf drop and stunting of the plant. Infection is worse during periods of extended rainfall, high humidity.


 Infected leaf part, mycelium and spores were observed under foldscope and following pictures were taken.

 We found some tetrad like structures while observing this sample and we couldn’t identify it.

 Antennae is serrate and dark in colour (Fig 1)

 Elytra with punctate distinct lines and gray spots (Fig 2)

 The femur of the hind leg are enlarged and bear a tooth and three tarsal segments  in hind leg (Fig 3)
 </li>
 <li>
 Hind wing with folding lines and has dot like cells between the lines(Fig 4)
 </li>
