Methodology:
10ml of human blood sample was collected and mixed well with glass beads for 30 minutes. 5 ml of Histopaque was placed in a 15 ml centrifuge tube. 1:1 ratio of Blood and PBS was carefully added onto the Histopaque-1077 Centrifuge the tubes for 30 min at 100 x g in 4 °C in a swing-out bucket. Aspirate the buffy coat formed in the interphase between histopaque and medium. Wash and centrifuge the cells for 100 x g for 10 min twice with 10 ml of PBS. The cells are then stained with Giesma Stain and observed under foldscope and microscope. PBS- Isotonic Phosphate Buffer Saline Solution
