<p>
 Methodology:
</p>
<ul>
 <li>
  A thin smear of blood was made on a clean slide and air- dried completely.
 </li>
 <li>
  Methanol was added and incubated at room temperature for 5 minutes.
 </li>
 <li>
  Once all methanol evaporates, add giemsa stain and incubate for 20 minutes. Wash in slow running tap water.
 </li>
 <li>
  Air dry the slide completely.
 </li>
 <li>
  Observe the slides under 10X Microscope and Foldscope.
 </li>

Differential Blood Staining

<p>
 Methodology:
</p>
<ul>
 <li>
  10ml of human blood sample was collected and mixed well with glass beads for 30 minutes.
 </li>
 <li>
  5 ml of Histopaque was placed in a 15 ml centrifuge tube.
 </li>
 <li>
  1:1 ratio of Blood and PBS was carefully added onto the Histopaque-1077
 </li>
 <li>
  Centrifuge the tubes for 30 min at 100
  <em>
   x g
  </em>
  in 4 °C in a swing-out bucket.
 </li>
 <li>
  Aspirate the buffy coat formed in the interphase between histopaque and medium.
 </li>
 <li>
  Wash and centrifuge the cells for 100 x g for 10 min twice with 10 ml of  PBS.
 </li>
 <li>
  The cells are then stained with Giesma Stain and observed under foldscope and microscope.
 </li>
</ul>

 PBS- Isotonic Phosphate Buffer Saline Solution
