 On both sides of leaf surface numerous small, superficial and blackish distinct tar like spots which  surrounded by yellow zone were observed. Along with the tar spots, there was light yellowish stroma and mats of hyphae of fungus were found on the upper surface of leaves. They were observed under foldscope and spores were found.

 It was tar leaf spots. This caused by
 
 Rhytisma acerinum
 
 fungus.

 The
mango leaf was found infected with fungal disease. The infected parts were
observed under foldscope. Literatures suggests that different kind of fungal
diseases like phoma blight, anthracnose, red rust, mango malformation, shooty
mould etc, may occur on mango leaf surface.


 This
kind of fungal infestation may cause loss of young shoots, flowers and limiting
fruit production. Spore production by these fungi is favored by wet or humid
weather.

Fungal infection of mango leaf

The spores on mango kernels are stained with lactophenol cotton blue solution and observed under foldscope. Lactophenol cotton blue stain is formulated with lactophenol, which serves as a mounting fluid, and cotton blue. Organisms suspended in the stain are killed due to the presence of phenol. Cotton blue is an acid dye that stains the chitin present in the cell walls of fungi.

Hands-on-training workshop was conducted on foldscope, at Hongirara school of Experience (Karnataka), as a part of Atal Tinkering Lab initiation, where the PUC students were taught about microscopy. The students was also educated about different microscopes for viewing microscopic structures. The students were trained on the usage of low-cost frugal paper microscope, i.e. the foldscope. During this half-day workshop the students were allowed to work with foldscope microscope, where they learnt the technique to prepare samples for viewing under foldscope device and also the technique to view as well as record the microstructure of the samples using foldscope.

Hands-on-training workshop conducted on Foldscope

Black cumin is an Indian spice, which was found infested with some kind of pests. A pest was captured and viewed under Foldscope.


 With the help of Foldscope, we were able to study the insect much closely and also viewed its larval form. I look forward to submitting my next post on the larva and eggs identification.

 We are viewing “Baker’s yeast” sample, using Foldscope, stained with Gram stain. We are able to get a clear vision of the yeast cells after gram staining.

Viewing Baker&#8217;s Yeast-#Indiafoldscopephase 1

Bacteria were isolated from soil collected from the nearby field. A known weight of soil sample was diluted in distilled water and the standard protocol was followed for pure culture isolation.


 The obtained pure culture was diluted with sterile water and gram staining was done using the diluted sample.


 After gram staining, the slide was observed under foldscope and violet coloured rod shape cells were found.

 Elytra have rows of pits giving them a striated (lined) appearance (Fig 2)

 Five segments in  tarsus of hind leg (Fig 3)

 Hind wing with folding lines and has dot like cells between the lines( Fig 4)
