Observation of the stomata of the leaf of guggul ( Commiphora wightii ) plant
The stomata are pores that are bordered by the guard cells and help the plant in gas exchange. A temporary mount of the leaf peel of the guggul ( Commiphora wightii ) plant was prepared and its stomata were observed as follows:
A fresh leaf was plucked from the guggul ( Commiphora wightii ) plant and was rinsed in the distilled water. The leaf was air-dried and was peeled from its center to expose the epidermis of the dorsal side (back side) of the leaf. A small piece of the peeled leaf was transferred to a watch glass containing the distilled water. Few drops of the safranin staining solution were then added to the watch glass and the leaf peel was stained for 30 seconds. The stained leaf peel was then transferred to another watch glass containing distilled water. A drop of glycerine was placed on a microscopic slide and the stained leaf peel was placed on it. A coverslip was placed over the specimen and any excess water/stain was removed using a blotting paper. The sides of the coverslip were attached to the microscopic slide with an adhesive tape. The leaf peel was observed in the compound microscope using the 10 X objective lens. The glass slide was placed in the foldscope and the stomata of the leaf peel were observed under the Foldscope. Also, the foldscope with the specimen was placed on the stage of the compound microscope and was observed using the microscope’s 10 X objective lens.
