A one-day event was conducted by Centre for Nanobiotechnology, VIT Vellore, on 16 March 2019 to aware UG/PG students and faculty members about the Foldscope and its potential advantages and applications.


 The event was started with the journey of the invention of Foldscope from match-stick model to paper-origami based microscope development followed by initiatives taken by DBT, Govt. of India, to bring the Foldscope in India.


 In the next session, all the participants assembled the Foldscope which was really unbelievable for them.


 Thereafter, three hours’ time task was given to all the participants to participate in the Image Acquisition contest where they were given the task to capture images/videos of any sample of their choice using Foldscope.


 The response of the contest was amazing and many participants could capture stunning videos and images during their first hands-on training of Foldscope. The images and videos were evaluated by experts, who are also faculty at VIT and based on their evaluation, contest results were declared.


 Overall, the experience of the participants was amazing. They loved the journey of Foldscope invention and they are eagerly waiting for the next workshop on Paper Centrifuge “Paperfuge”.

Imaging of Onion Peel Cells Through Foldscope

Salt-stress induced cell death in the unicellular green algae
 
 Chlorella
 
 vulgaris
 
 
 was investigated by incubating the algae cells with high salinity NaCl solution (150 mM) which typically causes cell damage or cell-death due to the osmotic potential which affects the permeability of the plasmalemma, cell growth and metabolic rate of the algae cells. The damaged cells can be characterized by the presence of non-uniform cell membrane while dead cells appear transparent due to leakage of chloroplast (without staining; encircled red ) or brown in color (after iodine staining; encircled yellow) as shown in the images captured using Foldscope. In contrast, the live cells appear green (or dark green in case of iodine staining) in color.

 are microscopic aquatic invertebrates that constitute the phylum Rotifera and found in aquatic environments all around the world

 The name Rotifer comes from the Latin words “wheel bearers” because the circular arrangement of moving cilia (tiny hairlike structures) at the front end resembles a rotating wheel.
 </li>
 <li>
 The cilia on the corona beat to create a vortex that draws in food for the rotifer to eat, which consist of dead organic matter, algae, protozoa and organisms small enough to fit in its mouth.
 </li>
 <li>
 In some species, the corona can also be used to propel the rotifer forward and help it swim.
 </li>
 <li>
 Sample Collection: The water sample was collected from the muddy puddle (stagnant water) within our campus.

Hexagonal Array Packing of Ommatidia in Mosquito Eye through Foldscope

<ul>
 <li>
 The solution of poly(ε-caprolactone) (PCL) of varying concentrations were prepared and used to fabricate nano- and micro-spun fibers for biomaterial applications. If any beads are present in these electrospun fibers (which is an indication of poor quality fibers), it suggests the need to optimize the parameters (such as temperature, humidity, concentration, speed, etc.). Here, the foldscope could be used to observe the presence of the beads in the electrospun fibers as encircled red in the image no.1. Further, the parameters were optimized accordingly which resulted in fibers without beads as shown in the image no. 2.
 </li>
 <li>
 In addition, foldscope could be used to determine the size of the electrospun fibers using a 200 µm core optical fiber as a reference tool to measure the dimension. The average diameter of the electrospun fibers was found to be 8.60±0.21 µm.
 </li>

Foldscope application in quality control of electrospun fibers by observing beads formation

 The blood smear was stained with Leishman stain and observed under the foldscope thereafter. The Leishman stain consists of two chemical groups, i.e. Azure B (basic dye) and Eosin Y (acidic dye). The erythrocytes stain pink in color due to Eosin Y as hemoglobin is eosinophilic (acidophilic), whereas the Azure B is a cationic dye which gives purple color to the nucleus and blue/light blue to the cytoplasm.
 </li>
 <li>
 This procedure and foldscopic exercise can be utilized to identify and differentiate leucocytes, malaria parasites, and Trypanosoma.
 </li>
</ul>

Observation of Stained Blood Smear Using Foldscope

<ul>
 <li>
 Human cheek cells can be observed under a microscope after staining with methylene blue dye. Since methylene blue (cationic in nature) binds with DNA and RNA (anionic in nature) by electrostatic means, it gives a darker blue color to nucleic materials as represented in the image given below.
 </li>
 <li>
 This procedure and foldscopic exercise can be used to observe any infection in the buccal cavity.
 </li>
 <li>
 Further, this can be used observe the presence of micronuclei in the buccal cells which are more in number in the smokers as reported in the literature (Proia et al 2006, Cancer Epidemiol Biomarkers Prev 2006;15(6). June 2006 1061-1077).
 </li>
</ul>
